ABSTRACT

Objective:To investigate the effect of milk on sebum secretion in golden hamsters, and to explore its possible mechanism of action.Methods:Eighteen golden hamsters were randomly and equally divided into 3 groups: blank control group receiving no intervention, whole-milk group gavaged with whole milk, and skimmed-milk group gavaged with skimmed milk. The gavage feeding was performed at a dose of 2.5 ml twice a day for 4 consecutive weeks. The maximum transverse diameter and maximum longitudinal diameter of bilateral sebaceous gland spots were measured on days 0, 7, 14, 21 and 28 after the start of intervention, and the area of sebaceous gland spots was calculated; at 24 hours after the last gavage, bilateral sebaceous gland spot tissues were resected, and subjected to immunohistochemical study to determine the expression of insulin-like growth factor-1 (IGF-1) /sterol regulatory element-binding protein-1 (SREBP-1) /acetyl-coenzyme A carboxylase (ACC-1) signaling pathway in sebaceous gland spots. Statistical analysis was carried out by using repeated measures analysis of variance, one-way analysis of variance for independent groups, Kruskal-Wallis H test, and least significant difference- t test for multiple comparisons. Results:Repeated measures analysis of variance showed that there was no significant difference in the area of sebaceous gland spots of golden hamsters among the 3 groups ( F= 0.96, P= 0.417) . The IGF-1 expression was significantly higher in the skimmed-milk group (0.39 ± 0.03) than in the blank control group (0.35 ± 0.03, t= 2.62, P= 0.021) and whole-milk group (0.33 ± 0.02, t= 3.82, P= 0.002) ; compared with the blank control group (0.36 ± 0.02) , the skimmed-milk group showed significantly increased SREBP-1 expression (0.42 ± 0.04, t= 2.64, P= 0.021) ; the ACC-1 expression was significantly higher in the skimmed-milk group (0.40 ± 0.03) and whole-milk group (0.40 ± 0.05) than in the blank control group (0.34 ± 0.03; t= 2.39, 2.47, P= 0.031, 0.026, respectively) . Conclusion:Milk may promote sebum secretion in golden hamsters through the IGF-1/SREBP-1/ACC-1 signaling pathway.

ABSTRACT

Objective:To evaluate the effect of pomegranate peel polyphenols on sebum secretion in golden hamsters, and to explore its possible mechanisms.Methods:Thirty golden hamsters were randomly and equally divided into 5 groups: (ointment) vehicle group, 0.48%-, 0.96%-, 1.92%-pomegranate peel polyphenol ointment groups, and retinoic acid cream group. Corresponding cream or ointments were applied to bilateral sebaceous gland spots of the golden hamsters at a dose of 1 gram twice a day for 4 consecutive weeks. The area of bilateral sebaceous gland spots was measured on days 0, 7, 14, 21 and 28 after the start of treatment, which was calculated by the maximum longitudinal diameter multiplied by the maximum transverse diameter. Twenty-four hours after the last treatment, immunohistochemical study was conducted to determine the expression of AKT/Sox9 signaling pathway in sebaceous gland spots resected from the golden hamsters. The area of sebaceous gland spots in these groups at different time points was compared by repeated measures analysis of variance, and other data were analyzed by one-way analysis of variance or Kruskal-Wallis H test. Results:The area of sebaceous gland spots was significantly smaller in the 0.96%-, 1.92%-pomegranate peel polyphenol ointment groups (50.48±2.41 mm 2, 48.24±2.56 mm 2, respectively) and retinoic acid cream group (48.31±2.76 mm 2) than in the vehicle group (57.99±3.29 mm 2; t=2.69, 3.98, 3.65, P=0.012, 0.001, 0.001, respectively) . Sox9 expression was significantly lower in the 1.92%-pomegranate peel polyphenol ointment group (0.39±0.04) and retinoic acid cream group (0.38±0.03) than in the vehicle group (0.44±0.02, P=0.040) . However, there was no significant difference in AKT expression among the 5 groups ( F=1.645, P=0.199) . Conclusion:Pomegranate peel polyphenols can reduce the sebaceous gland spot area and inhibit sebum secretion in golden hamsters, which may be related to the inhibition of Sox9 expression.

RESUMEN Objetivos. Identificar y determinar la estructura del fitoconstituyente de hojas de Tessaria integrifolia Ruiz & Pav con efecto leishmanicida. Materiales y métodos. Se preparó un extracto fluido de hojas de Tessaria integrifolia Ruiz & Pav. que fue concentrado a extracto blando y se utilizó para evaluar la actividad leishmanicida en Mesocricetus auratus con leishmaniasis experimental, administrando vía intramuscular la dosis de 250 mg/kg del extracto blando por 15 días. El extracto fue fraccionado en columna cromatográfica de 45 cm, con un diámetro de 2,5 cm que contiene silicagel G-60, de 70-230 mesh (Sigma-Aldrich®), las nueve fracciones obtenidas fueron evaluadas sobre macrófagos infectados con Leishmania sp para determinar la fracción activa y aislar el compuesto activo, mediante separación, purificación y cristalización, siendo analizado por resonancia magnética nuclear (RMN) de 1H, 13C, y cromatografía líquida acoplada a espectroscopía de masas (LC/MS). Resultados. El extracto fluido de las hojas de Tessaria integrifolia Ruiz & Pav. presenta actividad leishmanicida en Mesocricetus auratus y la fracción F8 es activa sobre macrófagos infectados a dosis de 14 µg/mL. Se elucidó en esa fracción un sesquiterpeno tipo eudesmano ((4aS, 5R,6R,8aR)-6-hidroxi-5,8a-dimetil-3-(1-metiletiliden) octahidronaftalen-2(1H)-ona), según análisis de RMN 1H, 13C, y LC/MS. Conclusiones. El extracto fluido de hojas de Tessaria integrifolia Ruiz & Pav. presenta actividad leishmanicida sobre Mesocricetus auratus. La fracción F8 presenta actividad leishmanicida sobre macrófagos infectados a dosis de 14 µg/mL. Se elucidó en la fracción activa un sesquiterpeno tipo eudesmano según los análisis de RMN 1H, 13C, y LC/MS.


ABSTRACT Objective. To identify and determine the phytoconstituent structure of Tessaria integrifolia Ruiz & Pav. leaves with leishmanicidal activity. Materials and Methods. Fluid extract of leaves was prepared, concentrated to soft extract, and used to evaluate leishmanicidal activity in Mesocricetus auratus with experimental leishmaniasis, at the dose of 250 mg/kg of soft extract by intramuscular route for 15 days. Extract was fractionated in 45 cm column chromatography with a 2.5 cm diameter, containing G-60 silica gel, and 70-230 mesh (Sigma-Aldrich®). Nine fractions were obtained and assessed on macrophages infected with Leishmania sp to determine the active fraction and isolate the active compound, by separation, purification, and crystallization, analyzed by nuclear magnetic resonance (NMR) of 1H, 13C, and liquid chromatography coupled to mass spectroscopy (LC/ MS). Results. Fluid extract from the leaves of T. integrifolia presents leishmanicidal activity in M. auratus. Fraction F8 is active on infected macrophages at a dose of 14 μg/mL. An eudesman type sesquiterpene ((4aS, 5R, 6R, 8aR) -6-hydroxy-5, 8a-dimethyl-3- (1-methylethylidene) octahydronaphthalen-2 (1H) -one) was identified, by RMN 1 H, 13C, and LC / MS analysis. Conclusions. Fluid extract of leaves of Tessaria integrifolia Ruiz & Pav. presents leishmanicidal activity on Mesocricetus auratus with experimental leishmaniasis. Fraction F8 presents leishmanicidal activity on infected macrophages at a dose of 14 μg/mL. An eudesman type sesquiterpene was identified, according to 1 H, 13C, and LC / MS NMR analysis.

Abstract Purpose: To evaluate the effects of red propolis on cheek pouch angiogenesis in a hamster new model sponge implant. Methods: Forty eight animals divided into eight groups. (Groups I-IV), the animals were treated for 15 days before and 10 days after sponge implantation. (Groups V-VIII), the animals were treated for 10 days after sponge implantation (GI and GV: red propolis 100 mg/kg, GII and GVI: celecoxib 20 mg/kg, GIII and GVII: 1% gum arabic 5 mL/kg, GIV and GVIII: distilled water 5 mL/kg). On the 11th day of implantation, the animals were anesthetized for stereoscopic microscopic imaging and morphometric quantification of angiogenesis (SQAN), followed by histopathological evaluation (H&E). Results: In the SQAN analysis, no significant difference was found between the groups. However, on histology, propolis was found reduce the population of mastocytes in the qualitative analyses (p = 0,013) in the quantitative analyses to reduce the number of blood vessels (p = 0,007), and increase the macrophage count (p = 0,001). Conclusion: Red propolis inhibited inflammatory angiogenesis when administered before andcontinuously after sponge implant, and was shown to have immunomodulating effects on inflammatory cells (mastocytes and macrophages) in a new sponge implant hamster model.

ABSTRACT

Hyperlipidemia is a condition of high lipid levels in the plasma and often linked with the deposition of lipid droplets in the aorta which initiate the progression of atherosclerosis. Atherosclerosis is a common cardiovascular disorder initiated by the formation of foams cells in the vascular wall which leads to turbulent blood flow, injury to the endothelial layer and subsequent vascular thrombosis. Since the early 1980’s, Golden-Syrian hamsters have been widely used as an animal model in the research of hyperlipidemia and atherosclerosis. The use of hamsters in the hyperlipidemic and atherosclerotic model is due to their lipoprotein profile that is closer to human setting, sensitive to high-fat high-cholesterol (HFHC) diet and a suitable rodent model. Atherosclerosis can be induced in hamsters through dietary challenge with HFHC diet. Over the decades, coconut oil (CNO) was commonly used as the source of fat in the diet design of high saturated fatty acids (SFA) composition. In this review, we summarized published literature with designs involving CNO plus cholesterol-induced hyperlipidemia, atherosclerosis or both. The factors that may influence the ability of CNO and cholesterol combination to induce hyperlipidemia such as the period of dietary intervention, hamster strains and the dietary amount were evaluated and summarized.

INTRODUÇÃO: Toxocaríase é uma infecção parasitária de distribuição global, causada pela fase larval de Toxocara spp. Os hospedeiros naturais são cães e gatos, nos quais o parasita completa o ciclo chegando a fase adulta. Outros hospedeiros podem ser infectados pela fase larval do parasita, após ingestão de ovos embrionados do solo, mãos contaminadas, fomites, ou ingestão de carne ou vísceras de animais infectados. Em hospedeiros paratênicos o parasita não completa o ciclo, invadindo em estágio larval vísceras ou outros tecidos, onde podem sobreviver e induzir a patologia. O presente estudo teve como objetivo caracterizar o hamster (Mesocricetus auratus), como modelo experimental de toxocaríase, inicialmente através do estudo das lesões histopatológicas em fígado, pulmão e rim. A caracterização da resposta imunológica do modelo, foi feita através do estudo de citocinas envolvidas nas respostas Th1 e Th2, e foi sugerida uma correlação entre alterações glomerulares e depósitos de complexos antígenos-anticorpo pré-formados na circulação. MÉTODOS: Hamsters foram inoculados com ovos embrionados de Toxocara canis, e mantidos no biotério do Instituto de Medicina Tropical de São Paulo. O estudo histopatológico foi desenvolvido utilizando-se cortes parafinados corados por hematoxilina e eosina. Para detecção de antígenos nos tecidos foram realizadas reações imunohistoquímicas, utilizando-se anticorpo monoclonal e policlonal anti- Toxocara canis. Utilizando-se o soro dos animais infectados e animais controle, foi realizada pesquisa de antígeno e anticorpo por ELISA. Para pesquisa de imunoglobulinas IgG e IgM e complemento, foram utilizados cortes congelados de rins para realização de reação de Imunofluorescência. INTRODUCTION: Toxocariasis is a parasitic infection of global distribution, caused by the larval stage of Toxocara spp. The natural hosts are dogs and cats, in which the parasite completes the cycle reaching adulthood. Other hosts can be infected with the larval stage of the parasite, after ingestion of embryonated eggs from the soil, contaminated hands, fomites, or ingestion of meat or viscera of infected animals. In paratenics hosts the parasite not complete the cycle, encroaching on larval stage in viscera or other tissues where they can survive and induce pathology. The present study aimed to characterize the hamster, Mesocricetus auratus, as experimental model of toxocariasis, initially through the study of histopathological lesions in the liver, lung and kidney. The characterization of immune response model, was made through the study of cytokines Th1 and Th2 responses involved, and a correlation was suggested between glomerular changes and antibody-antigen complexes deposits preformed in the circulation. METHODS: Hamsters were inoculated with embryonated eggs of Toxocara canis, and kept in the bioterium of the Institute of Tropical Medicine of the São Paulo. The histopathologic study was developed using paraffin slides stained by hematoxylin and eosin. For detection of antigens in tissues immunohistochemistry reactions were performed using monoclonal and polyclonal anti-Toxocara canis sera. Using the serum of infected and control animals, search has been carried out of antigen and antibody by ELISA. For the search of immunoglobulins IgG, IgM and complement, were used slides prepared from frozen fragments of kidneys and a immunofluorescence reaction. Fragments of kidneys were included for electron microscopy to detect antigens of Toxocara and immune complexes. Introducción. Es necesario desarrollar modelos de estudio de la leptospirosis. Objetivo. Genotipificar un aislamiento de Leptospira proveniente de una persona con síndrome de Weil y evaluar, con el modelo experimental en Mesocricetus auratus , su dinámica de infección. Materiales y métodos. Se hizo la genotipificación por análisis de las secuencias génicas rrs 16S y lipL32 . Se determinó la dosis letal media en hámster inoculada por vía intraperitoneal. Se identificaron los patrones de química clínica, la duración de la leptospiremia, la leptospiruria y la histopatología, comparados con el mismo modelo inoculado con la cepa de Leptospira interrogans (Fiocruz L1-130). Resultados. Mediante análisis molecular se determinó que el aislamiento correspondía a la especie patógena Leptospira santarosai . La bacteria se recuperó a partir de tejido de riñón y de pulmón, y se detectó por medio de PCR lipL32 en el tercer día después de la infección. La proteína C reactiva aumentó en el quinto día después de la infección (3,25 mg/dl; valor normal: 0,3 mg/dl) con una disminución en el día 18 (2,60 mg/dl; valor normal: 0,8 mg/dl). Los biomarcadores de urea mostraron alteraciones indicativas de falla renal aguda (día 5 después de la infección: 49,01 mg/dl y día 18: 53,71 mg/dl). La histopatología mostró neumonía intersticial con diferentes grados de hemorragia, así como nefritis intersticial. Conclusión. Se identificó la presencia de la especie L. santarosai con capacidad patógena comparable con la cepa Fiocruz L1-130 de L. interrogans , de reconocida virulencia y tropismo pulmonar, en cuanto a los aspectos histopatológicos de tropismo a pulmón y riñón. Nunca antes se había evaluado en un modelo experimental un aislamiento de origen local bajo estos criterios biológicos.


Introduction: Is necessary to develop models for the study of leptospirosis. Objective: To genotype a Colombian strain of Leptospira isolated from a human with Weil´s syndrome and to evaluate its infection dynamics in the hamster experimental model. Materials and methods: Genotyping was performed by amplification and sequence analysis of the rrs 16S and lipL32 genes. The median lethal dose was determined in intraperitoneally inoculated hamsters. The patterns of clinical chemistry, the duration of leptospiremia, leptospiruria and pathological findings were studied and compared in the same animal model infected with L. interrogans (Fiocruz L1-130). Results: Molecular typing revealed that the isolate corresponded to the pathogenic species L. santarosai, which was recovered from hamsters´ kidneys and lungs and detected by lipL32 PCR from day 3 post-infection in these organs. There was a marked increase of C-reactive protein in animals at day 5 post-infection (3.25 mg/dl; normal value: 0.3 mg/dl) with decreases by day 18 (2.60 mg/dl: normal value: 0.8 mg/dl). Biomarkers of urea showed changes consistent with possible renal acute failure (day 5 post-infection: 49.01 mg/dl and day 18 post-infection: 53.71 mg/dl). Histopathological changes included interstitial pneumonia with varying degrees of hemorrhage and interstitial nephritis. Conclusion: The pathogenic species L. santarosai was identified in Colombia. Its pathogenicity as determined by tropism to lung and kidney was comparable to that of L. interrogans Fiocruz L1-130, well known for its virulence and pulmonar tropism. The biological aspects studied here had never before been evaluated in an autochthonous isolate.

Objective To establish the genetic and biochemical loci in Mesocricetus auratus and its albino mua-tant.Methods Protein isozyme cellulose acetate electrophoresis was used to determine the genetic and biochemical loci in Mesocricetus auratus and its albino mutant, using the genetic and biochemical loci of mice and rats.Results 25 biochemi-cal markers of Mesocricetus auratus and albino mutant were established, and polymorphism of their genetic biochemical loci was analyzed.Conclusions Polymorphism of biochemical loci is present in Mesocricetus auratus.Some differences exist between the genetic biochemical loci of Mesocricetus auratus and their albino mutant.These results laid the foundation for further study on genetic mechanism of albino mutation in Mesocricetus auratus.

OBJETIVO: Avaliar o impacto na expressão AgNORs e apoptose na próstata do hamster-Mesocricetus auratus (hMa) submetido à aplicação de finasterida. MÉTODOS: Vinte roedores da espécie hMa (n=20), machos foram separados aleatoriamente em grupos de dez animais: grupo-Finasterida (n=10) e grupo-Controle (n=10). No grupo-finasterida foi administrado 7,14 ng/mL de finasterida, subcutâneo (SC), no dorso, três vezes por semana, por 90 dias. Foi avaliada a expressão AgNORs como marcador de proliferação celular e a apoptose como marcador de morte celular. RESULTADOS: A expressão de AgNORs foi menor no grupo-finasterida, 2,846±0,877 versus 3,68 ±1,07 grumos argilófilos por micrômetro ao quadrado (µm²) no grupo-controle, p= < 0,0001. A apoptose foi mais frequente no grupo-finasterida, 53,62±1,389 versus 14,76 ± 2,137 µm² no grupo-controle, p= 0,0408. CONCLUSÃO: Observou-se diminuição da expressão de AgNORs e promoção da apoptose na próstata dos roedores em estudo, que foram submetidos à aplicação de finasterida.

METHODS: Twenty male rodents of the species HMA (n = 20) were randomly assigned to groups of ten animals: Finasteride group (n = 10) and the Control group (n = 10). In the finasteride group 7.14 ng / mL finasteride was subcutaneously (SC) administered on the back of the animals three times a week for 90 days. AgNOR expression was evaluated as a marker of cell proliferation and apoptosis as a marker of cell death. RESULTS: The expression of AgNORs was lower in the finasteride group, 2.846 ± 0.877 vs. 3.68 ± 1.07 argyrophilic regions per square micrometer (µm2) in the control group, p = <0.0001. Apoptosis was more frequent in the finasteride group, 53.62 ± 1.389 versus 14.76 ± 2.13 per µm2 in the control group, p = 0.0408. CONCLUSION: We observed decreased expression of AgNORs and promotion of apoptosis in the prostate of rodents treated with finasteride.

ABSTRACT

PURPOSE: Evaluate the effects of finasteride on the serum PSA and on the prostate of hamster-Mesocricetus auratus(hMa). METHODS: Twenty hMa male adults were split in groups control and experimental (n=10). Animals of the experimental group received 7.14ng/mL of finasteride, subcutaneously (SC) on the back three times per week, during 90 days. The finasteride dose was equivalent to 5.0mg administered to a 70kg man. At the end of the experiment the mean age for the animals in the control group was 15.2±1.13months and for the experimental group was 17.7±0.67 months. There was a statistically significant difference between mean ages of both groups (t value=5.98; p=0.001). The animals of the control group weighted 129.0±18.8g and the experimental group weighted 145.0±15.5g, t=1.88 e p=0.0514. The serum PSA was assessed through ELISA method. Prostates of those animals were collected and processed to histology and morphometry: the diameter of the acinous glands and the acinous epithelium, apoptosis, AgNORs and cellularity were assessed in both groups. RESULTS: Serum PSA decreased in the experimental group, 0.003ng/mL versus 0.763ng/mL, H= 7.982 e p= 0.0047. Decrease in the acinous area occurred in animals that received finasteride, 238.000±24.600 μm² versus 398.600±55.320 μm²; t= 2.653; p= 0.0122. A remarkable decrease in the area of the acinous epithelium occurred in the animals that received finasteride, 111.900±12.820 μm² versus 160.400±18.430 μm² t= 2.162; p= 0.0361. OBJETIVO: Avaliar o efeito da finasterida no PSA sérico e na próstata do hamster-Mesocricetus auratus (hMa). MÉTODOS: 20 hMa adultos machos foram divididos em grupos de 10 animais. No experimento foram administrados 7,14 ng/mL de finasterida, subcutâneo (SC), no dorso, três vezes por semana, por 90 dias, dose equivalente a 5,0 mg usada em homem de 70Kg. Ao final da pesquisa, grupo experimento apresentou idade média de 17,7 ± 0,67 meses. O grupo controle apresentou idade média de 15,2 ± 1,13 meses. O valor de t na comparação das médias das idades entre os dois grupos foi de 5,98 e p=0.0001. Os animais-controle pesaram em média 129,0 ± 18,8g e o experimento 145,0 ± 15,5g; t=1,88 e p=0,0514. Na microscopia óptica de luz e estudo morfométrico: avaliaram-se o diâmetro dos ácinos e epitélio acinar prostáticos, a apoptose, a expressão AgNORs e a celularidade. RESULTADOS: O grupo-experimento apresentou média de PSA de 0,003 ng/mL e o grupo-controle de 0,763 ng/mL, H=7,982 e p=0,0047. A área dos ácinos do grupo-experimento foi de 238,000±24,600 μm² versus 398,600±55,320 μm²; t= 2,653; p= 0,0122. A área do epitélio acinar no grupo-experimento foi de 111,900±12,820 μm² versus 160,400±18,430 μm² t= 2,162; p= 0,0361. A expressão de AgNORs foi menor no grupo-experimento 2,846±0,877 versus 3,68 ±1,07 grumos argilófilos por μm², p= < 0,0001. A apoptose foi mais freqüente no grupo-experimento, 53,62±1,389 versus controle, 14,76 ± 2,137, p= 0,0408. Não houve diferença na celularidade entre os grupos de animais, 74,75±5,5 células no grupo-controle versus 65,07±13,24, no grupo-experimento, p= 0,5105. CONCLUSÕES: A finasterida diminuiu o PSA sérico, a área do lúmen, o epitélio acinar, a expressão de AgNORs e promoveu a apoptose nos ácinos da próstata dos hamsteres experimento e não houve diferença na celularidade acinar entre os animais estudados.

ABSTRACT

PURPOSE: To study the effect of finasteride on the spermatogenesis of adult Mesocricetus auratus. METHODS: Twenty adult hamsters were evaluated. The animals were one year-older, and were randomly divided in 2 different groups: control group with ten animals (n=10) and experimental group also with ten animals (n=10). The animals in the experimental group were shot 7.14 ng/mL (0.5mL) of finasteride by 100mg/Kg, subcutaneously in the dorsal region three times per week during 90 days. This dose correspondes to 5mg of the drug used in adult men for the treatment of benign prostatic hyperplasia (BPH). After three months, the animals were anesthetized through association of 200mg/kg ketamine chloridrate and 2.5 mg/kg of diazepan and were dead through hypovolemia.. The testis removed along with the whole genitourinary apparel were fixed with 10 percent formalin and submitted to histological analisys by optical microscopy. The hematoxilin-eosin (HE) method was used to stain the slides. RESULTS: The mean weight of animals in the control group before death was 129.0±18.8gr. The mean weight of animals in experimental group was 145.0±15.25gr. The mean age of animals in control group before death was 15.2±1.13 months. The mean age of animals in experimental group before death was 17.16±0.82 months. The mean difference in weight between both groups was not statistical significant (p=0.0514). The totality of animals in control group (100 percent) presented no tubular alterations and showed no disturbancy in the spermatogenesis stages. Four animals (40 percent) in the experimental group showed hypotrophy of the seminiferous tubules and six (60 percent) showed normal spermatogenesis, however reduced compared to control group. There was statiscally significant difference (p=0.043) between the control and experimental group related to testicular alterations. OBJETIVO: Estudar o impacto da finasterida na espermatogênese do Mesocricetus auratus, adulto. MÉTODOS: Foram avaliados 20 hamsters adultos, com idades superiores a 1 ano, distribuídos em dois grupos: grupo controle com dez animais (n=10) e grupo experimental também com dez animais (n=10). No grupo experimental foi aplicado 7,14ng/mL (0,5mL) de finasterida por 100mg/Kg/peso, subcutâneo (SC), na região dorsal do animal, três vezes por semana por 90 dias, dose correspondente a 5mg da droga usada em homens adultos, para tratamento da hiperplasia benigna da próstata (HBP). No final de três meses, esses Hamsters foram mortos por hipovolemia, após serem anestesiados com cloridrato de quetamina, na dosagem de 200 mg/kg juntamente com diazepam, na dosagem de 2,5 mg/kg. Os testículos foram retirados em monobloco juntamente com todo o aparelho geniturinário, fixados em formalina a 10 por cento e encaminhados à histotécnica para posterior análise histológica em microscópico óptico. Foram usadas para coloração das lâminas a hematoxilina e eosina (HE). RESULTADOS: Quando foram mortos, os Hamsters do grupo de controle pesaram em média 129,0g e desvio padrão (DP) de 18,8g. O grupo de experimento apresentou média de peso de 145,0g e DP de 15,25g. A idade dos animas de controle quando foram mortos apresentou média de 15,2 meses e DP de 1,13 meses. Os animais de experimento apresentaram média de idade de 17,16 meses e DP de 0,82. A diferença das médias de peso entre os dois grupos não teve significado estatístico (p=0,0514). Os animais (100 por cento) do grupo controle não tiveram alterações tubulares e apresentaram todas as etapas da espermatogênese normais. Quatro animais (40 por cento) do grupo de experimento apresentaram hipotrofia dos túbulos seminíferos, e seis (60 por cento) desses animais apresentaram espermatogênese normal, mas diminuída em relação ao grupo controle. ABSTRACT

A doença de Chagas, descrita há quase 100 anos por Carlos Chagas, continua sendo um problema de saúde pública na América Latina, com 15 milhões de infectados e 28 milhões de indivíduos susceptíveis. Vários mecanismos fisiopatológicos foram propostos para a progressão da miocardiopatia chagásica crônica, entre eles a denervação simpática. O uso do betabloqueador na insuficiência cardíaca congestiva diminuiu morbidade e mortalidade. Entretanto, nenhum trabalho conclusivo incluiu a miocardiopatia chagásica como etiologia. OBJETIVO: Avaliar o papel do carvedilol na sobrevida e remodelamento miocárdico na miocardiopatia chagásica experimental. MATERIAL E MÉTODOS: Foram avaliados 55 Hamsters Sirius, divididos em grupo controle, grupo infectado com Trypanosoma cruzi com 105 formas tripomastigotas via intraperitoneal e grupo infectado, tratado inicialmente com carvedilol 10 mg/Kg/dia e após 6 meses com 15 mg/Kg/dia em dose única por gavagem. Foi analisado o peso do animal; os diâmetros ventriculares, função sistólica e diastólica obtidas pelo ecocardiograma; a freqüência cardíaca, a duração do QRS, a presença de extra-sístoles e o ritmo cardíaco usando o eletrocardiograma e a avaliação da mortalidade. Após 12 meses os animais sobreviventes foram sacrificados e realizada a quantificação da fração do volume de colágeno intersticial e perivascular no miocárdio. RESULTADO: A razão dos diâmetros diastólico e sistólico pelo tamanho da tíbia não mostrou diferenças estatísticas, apesar de maiores nos grupos infectados. A fração de encurtamento também não mostrou diferença estatística, apesar de menor nos grupos infectados. A fração do volume de colágeno do ventrículo esquerdo e perivascular mostrou maior acúmulo nos grupos infectado e carvedilol em relação ao controle de forma significativa (p<0,001), mas sem diferença nos grupos infectados tratados ou não. A mortalidade foi maior nos grupos infectado e carvedilol (p = 0,001) e não ocorreu diferença entre estes grupos...


Chagas' disease was described 100 years ago by Carlos Chagas and nowadays it is a public health problem in Latin America yet. There are about 15 million infected people and another 28 million at risk to be infected. Several pathophysiologic mechanisms are suggested to be responsible for the progression of chronic Chagas' cardiomyopathy. Among them the sympathetic denervation seems to be an important issue. The use of beta blockade regarding heart failure has been proving to improve morbidity and mortality. However, none of these papers included Chagas' cardiomyopathy as etiology. OBJECTIVES: To evaluate the role of carvedilol upon survival and myocardial remodeling in a Chagas' cardiomyopathy animal model. MATERIAL AND METHODS: 55 Hamsters Sirius were studied and divided into control group, intraperitoneously infected group with Trypanosoma cruzi and infected group treated with carvedilol 10 mg/Kg/day by gavage which was increased to 15 mg/Kg/day after 6 months. Ventricular diameters, systolic and diastolic left ventricular function were evaluated by 2D-echocardiogram. The heart rate, QRS lasting, premature ventricular complex and cardiac rhythm were analyzed by ECG. We evaluated clinical parameters as the case for body weight. The survival curve was also studied. After 12 months the survivors were sacrificed and the myocardial interstitial and perivascular collagen volume fraction were analyzed. RESULTS: The ratio of diastolic or systolic diameters over tibia length did not show statistical differences although it was larger on infected groups. The fractional shortening also did not show statistical differences although it was decreased on infected groups. The left ventricular collagen volume fraction at the interstitial and perivascular area showed a higher accumulation on infected groups compared to control (p<0.001) but no differences were observed between infected and carvedilol treated animals. ABSTRACT

Syrian golden hamster (Mesocricetus auratus) has in its cheek pouches sub-epithelium an "Immunologically Privileged Site" which allows the integration of homo- and heterologous graft. This paper describes some anatomical and histological characteristics of that site, as well as analyzes aspects related with its immune properties. It also focuses the advantages of this experimental model over other models which are natural or induced carriers of immunodeficiency. Based on both these advantages and literature, this study aims to establish this model, through the performance of heterologous graft, as another option for the investigation of scar disturbances, as keloids and other diseases which may interest Plastic Surgery, as benign cutaneous lesions, and malignant neoplasias such as skin carcinomas and melanomas. The work also addresses perspectives for using this model, which still is a source scarcely known by Brazilian medical class.


O hamster sírio dourado (Mesocricetus auratus) possui no subepitélio de suas bolsas jugais um "Local de Privilégio Imunológico", permitindo a integração de homo e heteroenxertos. Este artigo descreve as características anatômicas e histológicas desse local, assim como os aspectos relativos as suas propriedades imunológicas. Cita, também, as vantagens desse modelo experimental sobre outros modelos portadores, naturais ou induzidos, de imunodeficiência. Com base nessas vantagens, e também baseado na literatura, tem o intuito de estabelecer esse modelo, mediante a realização de heteroenxertos, como uma outra opção para investigar distúrbios cicatriciais, como o quelóide, e doenças de interesse em Cirurgia Plástica, como lesões cutâneas benignas e neoplasias malignas como carcinomas e melanoma cutâneo. Tece, ainda, considerações sobre perspectivas de aplicações desse modelo, ainda bastante desconhecido entre a classe médica brasileira.

ABSTRACT

To describe the integration process of grafts of total human skin and keloid in hamster (Mesocricetus auratus) cheek pouch, whose sub-epithelium is naturally an "Immunologically Privileged Site". Fragments of human normal skin and keloid from the breast region of mulatto female patients were transplanted into the cheek pouch subepithelium in situ. Surgical procedure and grafted pouches for microscopic exam at several time points of the transplantation were standardized. The integration of grafted fragments of human skin and keloid was seen in late periods (84 days) since the microscopic assessment showed the presence of blood vases within the conjunctive tissue of grafted fragments. It was also possible to see among the grafted fragments the epithelium, the appearing of early cellular infiltrated, epithelial secretion of keratin, the presence of melanocytes, and delayed changes on the aspect of collagen fibers of conjunctive tissue. Pooled results allow to define hamster cheek pouch sub-epithelium as an experimental model to investigating heterologous graft physiology of human total skin and keloid with epithelium.


Descrever a integração dos enxertos de pele total humana e quelóide na bolsa jugal do hamster (Mesocricetus auratus), cujo subepitélio é, naturalmente, um "Local de Privilégio Imunológico". Foram transplantados fragmentos, de pele humana normal e de quelóide, obtidos da região mamária de pacientes pardas, no subepitélio da bolsa jugal in situ. O procedimento operatório, e de preparo das bolsas enxertadas para exame microscópico em vários períodos de transplante, foi padronizado. Verificou-se a integração dos fragmentos enxertados de pele humana e de quelóide em períodos tardios (84 dias), uma vez que a avaliação microscópica revelou a presença de vasos sangüíneos no tecido conjuntivo dos fragmentos enxertados. Foi também possível observar, nos fragmentos enxertados, o epitélio, o aparecimento de infiltrado celular incipiente, secreção epitelial de queratina, presença de melanócitos e alterações tardias no aspecto das fibras colágenas do tecido conjuntivo. Os resultados, em conjunto, permitem estabelecer o subepitélio da bolsa jugal do hamster como modelo experimental para investigação da fisiologia de tranplantes heterólogos de pele total humana e quelóide com epitélio.

ABSTRACT

Se caracterizaron microbiológicamente 2 cepas candidatas vacunales de Leptospira interrogans serogrupo Ballum, como parte de los trabajos encaminados a la obtención de nuevas formulaciones vacunales antileptospirósicas para uso humano. Se evaluó la cinética de crecimiento de ambas cepas en medio proteico EMJH y en 3 medios libres de proteínas. Se estimó la virulencia en hámsters mediante el cálculo de la dosis letal media. Fueron analizados los perfiles antigénicos celulares y extracelulares mediante SDS-PAGE unidimensional y comparados con aquellos de cepas pertenecientes a los serogrupos Canicola, Icterohaemorrhagiae y Pomona. La homología antigénica entre serogrupos heterólogos se analizó mediante western blotting con suero de hámsters vacunados con vax-SPIRAL®. Los resultados obtenidos demostraron un crecimiento fastidioso de ambas cepas de Ballum en los medios de cultivo estudiados, una alta virulencia en el modelo animal y una gran homología antigénica, con cepas pertenecientes a otros serogrupos de Leptospira de mayor circulación en Cuba.


Two candidate vaccines of Ballum serogroup Leptospira interrogans were microbiologically characterized as part of the work directed to the obtention of new antileptospirosis vaccine formulations for human use. The growth kynetics of both strains was evaluated in EMJH protein medium and in 3 protein free media. The virulence was estimated in hamsters by the calculation of the mean lethal dose. The cellular and extracellular antigenic profiles were analyzed by unidimensional SDS-PAGE and compared with those from strains of Canicola, Icterohaemorrhaagiaae and Pomona serogroups.The antigenic homology among heterologous groups was analyzed by western blotting with serun from hasterms vaccinated with vax-SPIRAL®. The results obtained showed a fastidious growth of both strains of Ballum in the studied media, a high virulence in the animal model and a large antigenic homology with strains from other serogroups of Lepstospira prevailing in Cuba.

ABSTRACT

Some aspects of the pathogeny of tuberculosis was experimentally investigated by inoculating either an attenuated mycobacteria strain BCG, or a virulent strainM. tuberculosis H37Rv into the foot pad or the cheek pouch of hamster. The inoculation of these agents determined different patterns of lesion. The BCG-induced lesionsin the foot pad had a tendency to descrease in size. On the other hand, the lesions induced by H37Rv showed a progressive evolution. The same pattern of evolution was observed with the lesions induced in the cheek pouch. However, the H37Rv disseminated to internal organs and the BCG did not. When BCG or H37Rv were inoculated in the foot pad of animals, the popliteal lymph node showed respectively a 21 fold and 68 fold increase in weight
